中国2型糖尿病遗传易感基因研究获重大进展--亲稳网络舆情监测室

  中国科学院上海生命科学研究院营养科学研究所研究员林旭主持的2型糖尿病基因组关联研究取得重大突破,首次发现了两个新的2型糖尿病易感基因位点,这两个位点的风险等位基因可能是分别通过损伤β细胞的功能和降低胰岛素的敏感性来增加2型糖尿病发病风险的。

  在该项目中,科学家不仅证实了23个在其他人群中报道的位点是中国人群的2型糖尿病易感基因位点,更为重要的是首次发现了两个新的2型糖尿病易感基因位点:RASGRP1-rs7403531和GRK5-rs10886471,而且通过基因功能分析,该项目还发现RASGRP1-rs7403531与胰岛中分泌胰岛素的β细胞受损相关,GRK5-rs10886471则与细胞对胰岛素的敏感程度相关。

  这些研究成果不仅有助于解析与中国人群2型糖尿病易感性相关的致病基因的结构、基因功能和生物学机理、种族间的差异,而且该项目所建立的人群数据库和生物样本库也将为揭示调控致病基因的主要营养和其他环境因素,为发展和建立对高危个体的预测模型和有效的干预方法提供研究平台,从而有助于建立符合中国人特点的2型糖尿病的早期风险评估和个体化干预,对遏制2型糖尿病在我国进一步蔓延有着重要意义。

  该项目为科技部863计划重点项目课题,林旭研究员率领的科研团队历时2年多展开科研攻关并取得了进展,合作单位包括中科院系统生物学重点实验室、北京大学人民医院、复旦大学附属华山医院、中国医学科学院基础医学研究所等。中国汉族人群的研究结果还在亚洲(AGEN-T2D)和欧洲(DIAGRAM)两个最大的全基因组关联研究合作联盟的人群数据库中进行了验证。
