中国科研人员发现精子细胞发育代谢调控机制--亲稳网络舆情监测室

   国际知名学术期刊Developmental Cell(《发育细胞》)1月15日发表了中科院上海生命科学院生物化学与细胞生物学研究所刘默芳组、王恩多组的合作研究成果,揭示了piRNA在精子发生后期触发MIWI/piRNA“机器”清除代谢调控机制,为代谢异常导致的男性不育症的发生提供了潜在的分子信息。

  研究组赵爽博士、苟兰涛博士及博士研究生张满等合作研究发现,在精子形成后期,piRNA分子通过与小鼠PIWI(MIWI)蛋白高度结合,促进其被泛素连接酶APC/C识别,从而使MIWI蛋白被泛素化修饰并降解;作为蛋白“配体”的piRNA自身也因失去结合蛋白MIWI的保护被机体清除,从而揭示了机体在生殖细胞发育特定阶段清除MIWI/piRNA“机器”的分子机制。更为重要的是,他们发现这种MIWI/piRNA“机器”的清除代谢是精子发育形成的必要条件,代谢异常将可能引起精子细胞发育受阻以致引起雄性或男性不育。(记者曹继军、颜维琦)
